Artificial metastases and decrease of fibrinolysis in the nude mouse lung after hemithoracic irradiation.

H Hirata, K Tanaka

    Clinical & Experimental Metastasis
    |October 1, 1984

    Summary

X-irradiation of the mouse lung promotes cancer metastasis. Radiation decreases fibrinolytic activity and natural killer cell function, creating conditions favorable for tumor spread.

    Area of Science:

    • Oncology
    • Radiology
    • Immunology

    Background:

    • Pulmonary metastasis is a significant challenge in cancer treatment.
    • The impact of localized radiation on the metastatic potential within the irradiated organ is not fully understood.

    Purpose of the Study:

    • To investigate the effect of X-irradiation on the nude mouse lung regarding the pulmonary metastasis of injected human cancer cells.
    • To explore the underlying mechanisms, including fibrinolytic activity and natural killer cell function.

    Main Methods:

    • Nude mice underwent right hemithoracic X-irradiation (20 Gy).
    • Human cancer cells were injected intravenously post-irradiation.
    • Metastasis, platelet aggregation, fibrin deposition, fibrinolytic activity, and natural killer cell activity were assessed.

    Related Experiment Videos

    Main Results:

    • Significant metastasis was observed in the irradiated right lung, with minimal metastasis in the non-irradiated left lung.
    • Increased platelet aggregation and fibrin deposition were noted around cancer cells in the irradiated lung.
    • Fibrinolytic activity and natural killer cell activity were reduced in the irradiated lung.

    Conclusions:

    • X-irradiation of the target organ, specifically the lung, enhances cancer cell metastasis.
    • Reduced fibrinolytic activity and natural killer cell function in the irradiated lung create a pro-metastatic microenvironment.
