    Area of Science:

    • Biochemistry
    • Analytical Chemistry
    • Clinical Diagnostics

    Background:

    • Accurate clinical analysis of heme precursors is crucial.
    • Routine porphyrin analysis requires an internal standard to validate procedural efficiency.

    Purpose of the Study:

    • To prepare and validate 2-Vinyl-4-hydroxymethyldeuteroporphyrin IX as an internal standard.
    • To demonstrate its utility in the analysis of porphyrins in biological samples.

    Main Methods:

    • Synthesis of 2-Vinyl-4-hydroxymethyldeuteroporphyrin IX.
    • Application of the internal standard in high-performance liquid chromatography (HPLC).
    • Resolution of uro- to protoporphyrins in urine samples.

    Main Results:

    • Successfully prepared 2-Vinyl-4-hydroxymethyldeuteroporphyrin IX.
    • Demonstrated its effectiveness as an internal standard for porphyrin quantification.
    • Validated the analytical method for normal and porphyric urine.

    Conclusions:

    • 2-Vinyl-4-hydroxymethyldeuteroporphyrin IX is a suitable internal standard for heme precursor analysis.
    • The developed method enhances the reliability of porphyrin measurements in clinical settings.
