    Summary

    1-methyl-3-phenylurea (MPU) can form the carcinogen 1-methyl-1-nitroso-3-phenylurea (MNPU) in the stomach. MNPU induced forestomach tumors in rats, suggesting a potential environmental cancer risk.

    Background:

    • 1-methyl-3-phenylurea (MPU) is a metabolite of the herbicide fenuron.
    • MPU itself is not carcinogenic.
    • Nitrosoureas are a class of compounds with known carcinogenic potential.

    Purpose of the Study:

    • To investigate the potential carcinogenicity of 1-methyl-1-nitroso-3-phenylurea (MNPU).
    • To determine if MNPU can be formed endogenously in rats.
    • To assess the role of MNPU as an environmental carcinogen.

    Main Methods:

    • In vitro formation of MNPU from MPU and sodium nitrite under acidic conditions.
    • Administration of MPU and sodium nitrite to rats to assess endogenous MNPU formation and carcinogenicity.
    • Histopathological examination of rat forestomach for tumor induction.

    Main Results:

    • MNPU was readily formed in vitro from MPU and sodium nitrite.
    • MNPU demonstrated significant carcinogenic activity in rats, inducing forestomach tumors.
    • Endogenous formation of MNPU was confirmed in rats co-administered MPU and nitrite, leading to tumor development.
    • MPU alone did not cause tumors.

    Conclusions:

    • Endogenous formation of MNPU from MPU and nitrite is a plausible mechanism for forestomach carcinogenesis.
    • MNPU represents a potential environmental carcinogen.
    • Findings have implications for preventive oncology and risk assessment of MPU-containing compounds.
