    Summary

    Dopaminergic regulation of prolactin is complex. Individual prolactin levels are stable over time, suggesting longitudinal studies are best for assessing mental illness. Combined drug challenges reveal more about dopamine abnormalities than single challenges.

    Background:

    • Dopaminergic (DA) transmission significantly influences pituitary prolactin (PRL) secretion.
    • Current methods assessing DA regulation in mental illness via PRL levels have limitations, including high interindividual variability and confounding factors.
    • Intraindividual variability of PRL levels and responses to DA challenges is not well understood.

    Purpose of the Study:

    • To investigate the stability of basal serum PRL levels in individuals over time.
    • To evaluate the utility of combined DA agonist and antagonist challenges for assessing DA regulation.
    • To determine if combined challenges offer greater insight into DA abnormalities compared to single challenges.

    • Combined DA agonist and antagonist challenges may be more effective in revealing DA dysregulation in abnormal populations than single-agent challenges.
