    Summary

This study compared dioxidine and gentamycin for treating Pseudomonas aeruginosa-induced pyelonephritis in rats. Both drugs showed similar effects on survival and kidney health, but dioxidine was more effective at reducing bacterial seeding from the kidneys.

    Area of Science:

    • Nephrology
    • Pharmacology
    • Microbiology

    Background:

    • Hematogenous pyelonephritis is a serious kidney infection.
    • Pseudomonas aeruginosa is a common causative agent of hospital-acquired infections.
    • Effective antimicrobial therapy is crucial for managing pyelonephritis.

    Purpose of the Study:

    • To evaluate the chemotherapeutic efficacy of dioxidine compared to gentamycin.
    • To assess the impact of these drugs on a rat model of Pseudomonas aeruginosa pyelonephritis.

    Main Methods:

    • A rat model of pyelonephritis was established using Pseudomonas aeruginosa.
    • Animals received intramuscular dioxidine (50-100 mg/kg/day) or gentamycin (5 mg/kg/day) for 7 days.
    • Outcomes measured included lethality, bacteriuria, kidney mass, macroscopic changes, and bacterial seeding from kidneys.

    Main Results:

    • Dioxidine and gentamycin demonstrated comparable effects on animal survival, bacteriuria, and kidney pathology.
    • Dioxidine showed a more significant reduction in bacterial seeding from the kidneys.
    • Neither drug achieved complete eradication of the infection.

    Conclusions:

    • Dioxidine exhibits comparable efficacy to gentamycin in treating experimental pyelonephritis.
    • Dioxidine may offer an advantage in reducing bacterial dissemination from the kidneys.
    • Further research is needed to optimize treatment regimens for complete recovery.
