    Background:

    • Lactoferrin, an iron-binding protein, is primarily found in secondary granules of polymorphonuclear leukocytes.
    • Its synthesis and accumulation patterns during granulocyte differentiation are not fully understood.

    Purpose of the Study:

    • To investigate the stages of lactoferrin biosynthesis during granulocyte maturation.
    • To determine if lactoferrin can serve as a marker for terminal granulocyte differentiation.

    Main Methods:

    • Examined lactoferrin biosynthesis in normal bone marrow and chronic myelogenous leukemia (CGL) samples.
    • Analyzed lactoferrin mRNA translation in Xenopus laevis oocytes using mRNA from CGL patients and HL-60 cells.

    Main Results:

    • Lactoferrin biosynthesis was absent in early myeloid cells but abundant in myelocytes and metamyelocytes.

    • The absence of lactoferrin in HL-60 cells is due to a lack of translatable mRNA, possibly indicating lineage infidelity or developmental arrest.
