    Summary

    Regulatory cells, including T cells and monocytes, are crucial for pokeweed mitogen (PWM)-stimulated B cell activation. Their influence on early B cell phospholipid synthesis is evident within 24 hours of in vitro culture.

    Background:

    • Pokeweed mitogen (PWM) stimulation of human B cells for DNA and immunoglobulin synthesis requires T cells and adherent cells.
    • The precise role of these regulatory cells in earlier B cell activation events remains unclear.

    Purpose of the Study:

    • To investigate the influence of T cells and monocytes on early B cell activation, specifically choline incorporation into phospholipids.
    • To determine the timing of regulatory cell influence on B cell responses to PWM.

    Main Methods:

    • Human B cells were cultured in vitro with or without PWM and T cells/monocytes.
    • Choline chloride ([methyl-14C]) was used to pulse cells at various time points to measure phospholipid synthesis.
    • B and T cells were separated to analyze their respective responses.

    Main Results:

    • PWM-induced phospholipid synthesis peaked in B cells at days 1-2, and in T cells at days 2-3.
    • In the initial 4 hours, monocytes showed the highest choline incorporation, followed by B cells, then T cells, with no PWM effect.
    • Between 16-20 hours, PWM significantly increased choline incorporation in B cells, dependent on T cells and monocytes.

    Conclusions:

    • The influence of regulatory T cells and monocytes on PWM-induced B cell activation is detectable within the first 24 hours of culture.
    • Early B cell phospholipid synthesis is modulated by the presence of T cells and monocytes during PWM stimulation.
