gamma-Glutamyl dopa: a kidney-specific dopamine precursor.

S Wilk, H Mizoguchi, M Orlowski

The Journal of Pharmacology and Experimental Therapeutics
    |July 1, 1978
    PubMed
    Summary

A novel gamma-glutamyl dopa pro-drug selectively generates dopamine in the kidney. This pro-drug significantly increases renal plasma flow, acting as a specific renal vasodilator with minimal systemic effects.

    Area of Science:

    • Biochemistry
    • Pharmacology
    • Nephrology

    Background:

    • Gamma-glutamyl derivatives are selectively taken up by kidney cells and metabolized.
    • Kidneys possess high concentrations of gamma-glutamyl transpeptidase and aromatic L-amino acid decarboxylase.
    • L-dopa is a precursor to dopamine, but its direct renal effects are limited.

    Purpose of the Study:

    • To synthesize and evaluate gamma-glutamyl dopa as a pro-drug for targeted renal dopamine delivery.
    • To investigate the efficacy of gamma-glutamyl dopa in increasing renal dopamine levels and renal plasma flow.
    • To assess the systemic side effects of gamma-glutamyl dopa compared to L-dopa.

    Main Methods:

    • Chemical and enzymatic synthesis of gamma-glutamyl dopa.
    • Administration of gamma-glutamyl dopa and L-dopa to mice and rats.
    • Measurement of renal dopamine concentrations and renal plasma flow.
    • Monitoring of systemic blood pressure (pressor effects).

    Main Results:

    • Gamma-glutamyl dopa administration resulted in a five-fold higher concentration of kidney dopamine compared to L-dopa.
    • A 10 nmol/g/30 min infusion of gamma-glutamyl dopa increased renal plasma flow by 60% in rats.
    • L-dopa at the same dose had no effect on renal plasma flow, and gamma-glutamyl dopa showed minimal pressor effects.

    Conclusions:

    • Gamma-glutamyl dopa serves as an effective pro-drug for selective dopamine generation within the kidney.
    • Gamma-glutamyl dopa demonstrates potential as a specific renal vasodilator.
    • The pro-drug approach offers targeted renal effects with reduced systemic impact.
