    Background:

    • Major affective disorder is a complex psychiatric condition with a suspected genetic component.
    • The Human Leukocyte Antigen (HLA) complex, located on chromosome 6, plays a crucial role in the immune system and has been investigated for its potential role in various diseases.
    • Previous hypotheses suggested a possible linkage between genes influencing affective disorder susceptibility and the HLA complex.

    Purpose of the Study:

    • To investigate the proposed genetic linkage between affective disorder susceptibility genes and the Human Leukocyte Antigen (HLA) complex.
    • To analyze Human Leukocyte Antigen (HLA) typing data from multiplex families with major affective disorder to test the linkage hypothesis.

    Main Methods:

    • Human Leukocyte Antigen (HLA) typing was performed on 15 unrelated multiplex families.
    • Data included 19 parents, 38 affected children, and 13 unaffected children.
    • Analysis focused on the distribution of shared HLA haplotypes among affected siblings, affected-unaffected siblings, and diagnostic subsets.

    Main Results:

    • The analysis of shared HLA haplotypes did not reveal any significant association.
    • No support was found for the hypothesis linking affective disorder susceptibility genes with the Human Leukocyte Antigen (HLA) complex on chromosome 6.
    • The distribution of haplotypes in affected and unaffected siblings was consistent with random inheritance.

    Conclusions:

    • The study fails to provide evidence supporting the proposed linkage between the Human Leukocyte Antigen (HLA) complex and major affective disorder.
    • Further research may be needed to explore other genetic or environmental factors contributing to the etiology of major affective disorder.
