Effects of gossypol on sperm counts in two inbred strains of mice
Abstract:
Gossypol acetic acid was administered orally to mice of two inbred strains, BALB/c/O1a and CBA/Gr, at daily doses of 10 or 20 mg/kg for about 4 weeks. Treated mice of both strains showed a reduction in sperm counts. This was more marked in CBA mice, which also had smaller testes size than did BALB/c mice. The treatment had no significant effect on testicular weight but the caput epididymidis and seminal vesicles of treated mice weighed less than those of control mice.
Insights
Gossypol acetic acid reduced sperm counts in mice. This male contraceptive effect was more pronounced in CBA mice, which also exhibited smaller testes and accessory sex organ weights.
Area of Science:
- Reproductive biology
- Toxicology
- Pharmacology
Background:
- Gossypol, a natural compound from cotton seeds, has shown antifertility effects in males.
- Understanding its impact on different mouse strains is crucial for assessing its therapeutic potential and risks.
Purpose of the Study:
- To investigate the effects of gossypol acetic acid on sperm count and reproductive organ weights in two inbred mouse strains.
- To compare the sensitivity of BALB/c and CBA mice to gossypol acetic acid treatment.
Main Methods:
- Oral administration of gossypol acetic acid (10 or 20 mg/kg daily) to BALB/c and CBA mice for approximately 4 weeks.
- Assessment of sperm counts and weights of testes, caput epididymidis, and seminal vesicles.
Main Results:
- Gossypol acetic acid treatment led to reduced sperm counts in both mouse strains.
- CBA mice showed a more significant reduction in sperm count and smaller testes size compared to BALB/c mice.
- While testicular weight remained unaffected, caput epididymidis and seminal vesicle weights were reduced in treated mice.
Conclusions:
- Gossypol acetic acid exhibits antifertility effects in male mice, evidenced by decreased sperm counts.
- Strain-dependent differences in response to gossypol acetic acid were observed, with CBA mice being more susceptible.
- Gossypol acetic acid may impact accessory sex organ function in addition to spermatogenesis.
