    Summary

    Ectopic calcified nodules from calcinosis interstitialis did not activate phagocytes in vitro. This suggests a mechanism for the lack of inflammation observed around these nodules.

    Background:

    • Recent studies show crystal phagocytosis stimulates the respiratory burst in human neutrophils.
    • Calcinosis interstitialis involves ectopic calcified nodules, but their interaction with immune cells is unclear.

    Observation:

    • Ectopic calcified nodules from calcinosis interstitialis were incubated with autologous granulocytes and monocytes.
    • Respiratory burst activity was measured using luminol-enhanced chemiluminescence.
    • Controls included various crystals (monosodium urate, hydroxyapatite) and zymosan.

    Findings:

    • Microcrystalline monosodium urate and hydroxyapatite (in granulocytes) strongly stimulated the respiratory burst.
    • Hydroxyapatite showed selective functional modulation, not stimulating monocytes.
    • Nodule material induced only marginal stimulation of phagocytes, unlike zymosan.

    Implications:

    • The limited stimulatory capacity of calcinosis nodule material may explain the absence of inflammatory reactions.
    • Understanding phagocyte-crystal interactions is crucial for inflammatory disease research.
    • This study provides insights into the pathobiology of calcinosis interstitialis.
