    Bile duct ligation in rats decreased smooth muscle responses to noradrenaline. Bile salts also reduced muscle contractility, but not enough to explain hypotension in obstructive jaundice.

    Background:

    • Obstructive jaundice is associated with reduced total peripheral resistance and hypotension.
    • The role of smooth muscle dysfunction in this condition is not fully understood.

    Purpose of the Study:

    • To investigate the impact of bile duct ligation on vascular and extravascular smooth muscle responsiveness.
    • To determine if bile salts possess spasmolytic properties that could explain hypotension in obstructive jaundice.

    Main Methods:

    • Isolated rat hindlimb perfusion was used to assess vascular smooth muscle.
    • Portal vein and vas deferens preparations examined extravascular smooth muscle responses.
    • Contractile responses to noradrenaline and tyramine were measured before and after bile duct ligation.

    Main Results:

    • Bile duct ligation significantly reduced contractile responses to noradrenaline in both vascular and extravascular smooth muscle.
    • Exposure to certain bile salts in vitro resulted in decreased smooth muscle contractility.
    • The spasmolytic effect of bile salts was dependent on the type and concentration of the bile salt.

    Conclusions:

    • The reduced responsiveness of smooth muscle to noradrenaline after bile duct ligation may contribute to hypotension in obstructive jaundice.
    • In vitro findings suggest that the hypotension observed in obstructive jaundice is unlikely to be solely due to a spasmolytic effect of bile salts on smooth muscle.
