    Background:

    • Investigating the mechanisms that regulate DNA replication.

    Purpose of the Study:

    • To determine if Xenopus laevis egg cytoplasm can initiate and sustain DNA replication in nuclei from Drosophila melanogaster.
    • To characterize the nature of DNA synthesis initiated in these reconstituted nuclei.

    Main Methods:

    • Isolation of nuclei from a permanent Drosophila melanogaster cell line.
    • Microinjection of nuclei and radioactive DNA precursors ([3H]TTP) into Xenopus laevis eggs.
    • Autoradiography to detect DNA synthesis.
    • Density labeling with BrdUTP followed by cesium chloride gradient centrifugation to analyze replication mode.

    Main Results:

    • In vitro cultured Drosophila nuclei showed minimal S phase activity (<7%).
    • Following injection into Xenopus eggs, 99% of nuclei synthesized DNA within 90 minutes.
    • Cesium chloride gradient analysis confirmed that the DNA synthesis was semi-conservative.

    Conclusions:

    • Xenopus egg cytoplasm contains signals capable of initiating and sustaining DNA replication.
    • These signals can effectively drive true semi-conservative replication in nuclei from an evolutionarily distant organism (Drosophila).
    • This highlights a conserved nature of fundamental DNA replication machinery and regulatory signals between amphibians and insects.
