    Summary

    Mutations in the H-2Kb locus of C57BL/6 mice did not affect immune response (Ir) gene function for tested antigens. This suggests the H-2Kb locus is not involved in Ir gene function for these specific immune responses.

    Background:

    • Immune response (Ir) genes regulate the ability of an organism to mount an antibody response to specific antigens.
    • The H-2 complex in mice, analogous to the human leukocyte antigen (HLA) complex, plays a crucial role in immune regulation and antigen presentation.
    • Mutations within specific loci of the H-2 complex can alter immune function, providing insights into gene mapping and Ir gene localization.

    Purpose of the Study:

    • To investigate the role of the H-2Kb locus in immune response (Ir) gene function.
    • To determine if mutations in the H-2Kb locus affect antibody responses to specific antigens.
    • To assess the impact of H-2Ld locus mutations on immune responses.

    Main Methods:

    • Studied four C57BL/6 mutant strains with presumed H-2Kb locus mutations.
    • Assessed antibody responses to (TG)-A-L, ovalbumin (OA), and lactic dehydrogenase B (LDHB) antigens.
    • Compared immune responses of mutant strains to parental C57BL/6 and BALB/cKh strains, including responses to the H-Y antigen.

    • Immune response gene mapping data supports the localization of these Ir genes outside the H-2Kb locus.
    • The BALB/c-dm2 mutation in the H-2Ld locus did not alter immune responses compared to the parental strain.
