    Summary

    This study shows that most Immunoglobulin A (IgA) in bile is secretory IgA, involving the secretory component (SC). Biliary epithelial cells, not liver cells, mediate IgA transfer.

    Background:

    • Immunoglobulin A (IgA) is crucial for mucosal immunity.
    • Secretory component (SC) facilitates IgA transport across epithelial barriers.
    • The role of IgA and SC in hepatic bile requires further elucidation.

    Purpose of the Study:

    • To quantify IgA in hepatic bile.
    • To characterize the association of IgA with secretory component (SC).
    • To investigate the cellular mechanisms of IgA transport in the biliary system.

    Main Methods:

    • Radioimmunoassay was used to measure IgA concentration in bile from 6 patients.
    • Immunochemical methods characterized IgA and SC.
    • Immunocytochemistry (peroxidase-labeled antibody method) localized IgA and SC in liver and bile duct tissues.

    Main Results:

    • Hepatic bile IgA concentrations ranged from 0.14 to 0.88 mg/ml.
    • Secretory IgA (IgA associated with SC) constituted 72% to 95% of total IgA.
    • Immunocytochemistry revealed IgA and SC in biliary epithelium, suggesting SC-mediated endocytic transfer.

    Conclusions:

    • Hepatic bile contains significant levels of secretory IgA.
    • Biliary epithelial cells, both intrahepatic and extrahepatic, are involved in the endocytic, SC-mediated transport of IgA.
    • Hepatocytes do not appear to participate in this IgA transport mechanism.
