    Background:

    • Glycosaminoglycans (GAGs) are crucial biomolecules with diverse biological roles.
    • Dermatan sulfates are a class of GAGs found in various tissues.
    • Understanding GAG structure is key to elucidating their functions.

    Purpose of the Study:

    • To characterize the peptide-linked dermatan polysulfates from hagfish and shark.
    • To investigate the glycosidic linkages between GAGs and peptides.
    • To identify novel structural features in these marine-derived GAGs.

    Main Methods:

    • Isolation and purification of dermatan polysulfate peptides from hagfish notochord, hagfish skin, and shark skin.
    • Analysis of amino acid and sugar composition.
    • Beta-elimination-reduction followed by analysis of released products to determine linkage types.

    Main Results:

    • Dermatan polysulfate peptides II and III showed O-glycosidic linkages between xylose and serine, similar to known chondroitin sulfates and dermatan sulfate.

    • The molar ratio of serine, xylose, and galactose was consistent across all three peptide types.

    Conclusions:

    • The linkage region of dermatan polysulfate peptide I represents a novel structure with dual O-glycosidic bonds involving N-acetylgalactosamine.
    • This finding is the first to report N-acetylgalactosamine in the linkage region of uronic acid-containing glycosaminoglycans.
    • The structural diversity of GAGs in marine organisms like hagfish and sharks is highlighted.
