Adenosine production in the ischemic kidney.

W L Miller, R A Thomas, R M Berne

    Summary

The ischemic kidney produces adenosine, a substance that may worsen kidney ischemia after blood flow is restored. This study identifies the proximal tubule as the primary site of adenosine production in the kidney.

    Area of Science:

    • Nephrology
    • Renal Physiology
    • Biochemistry

    Background:

    • Renal ischemia can lead to kidney damage.
    • Adenosine is a molecule known to affect blood vessel constriction.
    • The role of adenosine in kidney ischemia is not fully understood.

    Purpose of the Study:

    • To measure adenosine levels in the kidney, blood, and urine during renal artery occlusion.
    • To identify where in the kidney adenosine is produced and released.
    • To investigate the potential role of adenosine in post-ischemic kidney injury.

    Main Methods:

    • Renal artery occlusion in dogs.
    • Measurement of adenosine, inosine, and hypoxanthine in urine, renal tissue, and renal venous plasma.
    • Modified stop-flow experiments to localize adenosine in renal tubules.
    • Enzyme activity assays for 5'-nucleotidase in kidney cells.

    Main Results:

    • Urine and renal tissue concentrations of adenosine, inosine, and hypoxanthine increased after renal artery occlusion.
    • Elevated inosine and hypoxanthine levels were observed in renal venous plasma.
    • Adenosine was detected in proximal tubule fluid.
    • 5'-Nucleotidase, an enzyme involved in adenosine production, was localized to proximal tubule cells.

    Conclusions:

    • The ischemic kidney produces and releases adenosine.
    • Adenosine production is primarily associated with proximal tubule cells.
    • Adenosine may play a role in mediating renal ischemia following arterial occlusion.
