    Summary

    Germ-free dogs showed no fluid secretion above intestinal obstructions, unlike conventional dogs. This suggests gut bacteria, not the obstruction itself, cause fluid secretion in normal animals.

    Background:

    • Intestinal obstructions in conventional animals lead to fluid secretion above the blockage.
    • The role of gut microbiota in this physiological response is not fully understood.

    Purpose of the Study:

    • To investigate the role of gut bacteria in fluid secretion and mucosal changes above intestinal occlusions.
    • To compare the effects of mechanical obstruction in germ-free versus conventional dogs.

    Main Methods:

    • Mechanical intestinal occlusions were created in germ-free dogs.
    • In vitro analysis of intestinal loops included nutrient uptake (phenylalanine, beta-methyl-glucoside), influx kinetics, and mucosal morphometry.
    • Comparisons were made between loops above, below, and a control loop, as well as with data from conventional dogs.

    Main Results:

    • Intestine segments above the occlusion in germ-free dogs did not secrete water or electrolytes.
    • Transport properties and mucosal structure above the occlusion showed minimal changes compared to controls.
    • The segment below the obstruction exhibited significant atrophy, similar to conventional dogs.

    Conclusions:

    • The copious fluid secretion observed above intestinal obstructions in conventional animals is likely due to the presence of gut bacteria.
    • Germ-free conditions prevent the characteristic fluid secretion response to intestinal obstruction.
