Colchicine and insulin secretion in man.

D Giugliano, T Cerciello, N Passariello

    Summary

Colchicine administration, both acute and chronic, impairs human insulin secretion and glucose tolerance. Lysine acetylsalicylate (LAS) administration reversed these negative effects, suggesting a role for prostaglandin synthesis.

    Area of Science:

    • Endocrinology
    • Pharmacology

    Background:

    • Colchicine is used to treat gout and other inflammatory conditions.
    • Its effects on glucose metabolism and insulin secretion are not fully understood.

    Purpose of the Study:

    • To investigate the impact of acute and chronic colchicine administration on insulin secretion and glucose tolerance in humans.
    • To explore the role of prostaglandin synthesis in mediating these effects.

    Main Methods:

    • Acute intravenous colchicine administration and assessment of insulin response to glucose.
    • Chronic oral colchicine administration over 10 days with oral glucose tolerance testing.
    • Administration of lysine acetylsalicylate (LAS) to inhibit prostaglandin synthesis.

    Main Results:

    Related Experiment Videos

    • Acute colchicine significantly decreased insulin response to glucose and reduced glucose disappearance rates.
    • Chronic colchicine suppressed insulin secretion and worsened glucose tolerance.
    • LAS completely reversed the inhibitory effects of colchicine on insulin secretion and glucose tolerance.

    Conclusions:

    • Colchicine administration inhibits glucose-induced insulin secretion and impairs glucose tolerance in humans.
    • Increased prostaglandin E (PGE) synthesis may mediate colchicine's inhibitory effects.
    • LAS can counteract the negative metabolic effects of colchicine.
