    Summary

    Calcitonin (CT) administration in rabbits during saline diuresis significantly increased urine output and electrolyte excretion, primarily by reducing proximal sodium reabsorption. These findings suggest CT possesses diuretic and saliuretic properties.

    Background:

    • Calcitonin (CT) is a hormone with known effects on calcium and phosphate metabolism.
    • Its direct renal effects, particularly its diuretic and natriuretic potential, require further elucidation.

    Purpose of the Study:

    • To investigate the renal effects of porcine Calcitonin (CT) in rabbits.
    • To determine the impact of CT on water, electrolyte, calcium, and phosphorus excretion during saline diuresis.

    Main Methods:

    • Rabbits underwent isotonic (ISD) and hypertonic (HSD) saline diuresis.
    • Porcine CT was administered at various doses (0.1–20 IU MRC/kg/hr).
    • Excreted fractions of water, electrolytes (Na+, Cl-, K+), calcium, phosphorus, and osmolality were measured.

    Main Results:

    • During ISD, CT (4 and 20 IU MRC/kg/hr) significantly increased diuresis and excretion of Na+, Cl-, K+, PO4 3-, and Ca2+ in a dose-dependent manner.
    • A biphasic pattern in free-water clearance was observed during ISD.
    • During HSD, CT reduced free-water reabsorption and the urinary to plasma osmolality ratio.

    Conclusions:

    • CT exhibits diuretic and saliuretic effects in rabbits, primarily mediated by decreased proximal sodium reabsorption.
    • The findings suggest a potential role for CT as a diuretic agent, with its effects comparable to established diuretics.
