    This study reveals a two-phase mechanism for chemical carcinogenesis in hamsters. Early exposure to 7,12-dimethylbenz(a)anthracene (DMBA) followed by a later, stronger dose initiated tumor formation, suggesting distinct initiation and promotion phases.

    Purpose of the Study:

    • To elucidate the roles of initiation and promotion in tumor development using 7,12-dimethylbenz(a)anthracene (DMBA) in a hamster buccal pouch model.

    Main Methods:

    • Hamster buccal pouches were painted with varying concentrations and durations of 7,12-dimethylbenz(a)anthracene (DMBA).
    • Group 1: 0.1% DMBA for 10 weeks (no tumors).
    • Group 2: 0.1% DMBA for 10 weeks, followed by 6 weeks of no treatment, then 0.5% DMBA for 4 weeks (epidermoid carcinomas).
    • Group 3: 0.5% DMBA for 4 weeks (no tumors).

    • Prolonged exposure to a low concentration of DMBA (0.1%) did not induce tumors or dysplasia.
