    Summary

    This study visualizes widespread Leu-enkephalin and BAM-22P systems in rat brain regions using immunocytochemistry. Results suggest similar enkephalin biosynthesis mechanisms in brain and adrenal cells.

    Background:

    • Enkephalins are known neuropeptides involved in pain modulation and reward pathways.
    • Previous studies localized enkephalins to telencephalic neurons, but their distribution was not fully elucidated.

    Purpose of the Study:

    • To comparatively map the distribution of Leu-enkephalin and its precursor fragment, BAM-22P, in the rat telencephalon.
    • To visualize widespread enkephalin-containing neuronal systems in cortical-limbic regions.

    Main Methods:

    • Immunocytochemical procedures were employed to detect Leu-enkephalin and BAM-22P.
    • Rats were treated intraventricularly with colchicine to enhance neuronal perikarya visualization.

    Main Results:

    • Widespread Leu-enkephalin immunoreactivity was observed in neuronal perikarya and processes across numerous telencephalic regions.
    • BAM-22P immunoreactivity was found in the same regions, co-localizing with Leu-enkephalin.
    • Enkephalin systems were visualized in olfactory bulb, basal ganglia, amygdala, hippocampus, and various cortical areas.

    Conclusions:

    • The study successfully visualized extensive enkephalin-containing neuronal networks in the rat telencephalon.
    • Co-localization of Leu-enkephalin and BAM-22P suggests shared biosynthetic pathways.
    • Findings indicate that brain enkephalin neurons may utilize biosynthesis mechanisms akin to those in adrenal medullary cells.
