    As myocardial cells age, their DNA synthesis decreases, correlating with the appearance of specific non-histone nuclear proteins in rat hearts. These protein changes are linked to cellular aging and nuclear function.

    Background:

    Purpose of the Study:

    • To investigate the relationship between DNA synthetic activity and non-histone nuclear proteins in aging rat myocardial cells.
    • To characterize the differential expression of these proteins during postnatal development.

    Main Methods:

    • Isolation of myocardial cell nuclei from rats of various ages (4 days to 15 months).
    • In vitro assessment of DNA synthetic activity.
    • Analysis of non-histone nuclear protein profiles using isoelectric focusing (pI) and molecular weight (Mr).

    • Group c (pI 6.5-8.5, Mr 32000-42000) was present throughout, but decreased in older rats.

    Conclusions:

    • The age-related decline in myocardial cell DNA synthetic activity is associated with specific changes in non-histone nuclear protein composition.
    • These proteins are nuclear-localized and their differential expression may play a role in regulating nuclear function during cardiac aging.
    • Further analysis of amino acid composition provides insights into the nature of these age-associated nuclear proteins.
