    The spleen may cause age-related thymus shrinkage in rats. Splenectomy prevented thymus weight loss, suggesting the spleen influences thymic involution over time.

    Background:

    • Thymic involution, the age-related decrease in thymus size and function, is a hallmark of aging.
    • The spleen's role in thymic involution remains incompletely understood.

    Purpose of the Study:

    • To investigate the spleen's potential causative role in age-related thymic involution in young female rats.

    Main Methods:

    • Forty young female rats underwent splenectomy (n=20) or served as intact controls (n=20).
    • Body weight, thymus weight, and spleen weight were monitored monthly for up to one year.
    • Histological analysis was performed at 4 and 12 months.

    Main Results:

    • All rats exhibited a declining thymic index due to normal weight gain.
    • Splenectomized rats showed no significant change in absolute thymus weight.
    • Intact control rats displayed significantly reduced thymus weight at 4 and 12 months (P < .001 and P < .005, respectively).

    Conclusions:

    • The spleen appears to be a contributing factor in the gradual thymic involution associated with aging.
    • This contrasts with the rapid thymus weight loss observed during tumor growth.
