Analysis of pleural effusions using automated flow cytometry.

K M Unger, M Raber, C W Bedrossian

    Summary

Flow cytometry accurately analyzes deoxyribonucleic acid (DNA) content in cells. DNA analysis of pleural effusions shows hyperdiploid cells strongly suggest malignancy, aiding cancer diagnosis.

    Area of Science:

    • Oncology
    • Cell Biology
    • Medical Diagnostics

    Background:

    • Aneuploidy in solid tumors correlates with neoplastic cells.
    • Flow cytometry enables rapid, accurate DNA content analysis.
    • Pleural effusions require diagnostic evaluation for malignancy.

    Purpose of the Study:

    • To evaluate the utility of flow cytometry for DNA analysis in pleural effusions.
    • To compare flow cytometry DNA analysis with cytologic examination for malignancy detection.
    • To assess the diagnostic accuracy of DNA content abnormalities in pleural effusions.

    Main Methods:

    • Performed cytologic examination and DNA analysis by flow cytometry on pleural effusions from 33 patients.
    • Analyzed deoxyribonucleic acid (DNA) content for aneuploidy.

    Related Experiment Videos

  • Correlated flow cytometry results with cytologic findings and clinical diagnoses.
  • Main Results:

    • Flow cytometry and cytology agreed in 10 of 12 malignant and 20 of 21 benign effusions.
    • Two malignant effusions were false-negatives by flow cytometry.
    • One benign effusion showed hypodiploid cells (false-positive); hyperdiploid cells were highly suggestive of malignancy.

    Conclusions:

    • Flow cytometry DNA analysis is rapid, specific, and sensitive for detecting malignancy in pleural effusions.
    • Hyperdiploid cells in pleural effusions are a strong indicator of malignancy.
    • Flow cytometry complements cytologic examination in diagnosing malignant pleural effusions.
