    Area of Science:

    • Pharmacokinetics
    • Drug Metabolism
    • Clinical Pharmacology

    Background:

    • Amrinone is a cardiotonic agent.
    • Understanding amrinone's pharmacokinetic profile is crucial for safe and effective dosing.

    Purpose of the Study:

    • To evaluate the pharmacokinetics of amrinone in healthy subjects.
    • To determine the dose-proportionality and elimination characteristics of amrinone.

    Main Methods:

    • A randomized crossover study involving 18 healthy subjects.
    • Administration of amrinone at 75 mg, 150 mg, and 225 mg doses.
    • Measurement of plasma amrinone concentrations and calculation of pharmacokinetic parameters.

    Main Results:

    • Plasma amrinone levels rose proportionally to the administered dose.
    • Mean plasma AUC (area under the curve) values were 4, 8.18, and 12.35 µg·hr/mL for the respective doses.
    • Mean maximum plasma concentrations (Cmax) were 1.03, 1.74, and 2.58 µg/mL.
    • The elimination half-life (t1/2) was consistently around 3.85 hours and independent of dose.

    Conclusions:

    • Amrinone exhibits dose-proportional pharmacokinetics in healthy subjects within the studied dose range.
    • The elimination of amrinone is not dose-dependent, suggesting a linear pharmacokinetic model.
    • These findings support the predictable behavior of amrinone in terms of plasma concentration and elimination.
