    Background:

    • Prenatal alcohol exposure is a leading cause of preventable birth defects.
    • Fetal Alcohol Spectrum Disorders (FASD) encompass a range of lifelong physical, behavioral, and intellectual disabilities.

    Purpose of the Study:

    • To investigate the long-term behavioral and neuroanatomical consequences of prenatal alcohol exposure in a rat model.
    • To explore the relationship between learning deficits and hippocampal structural changes.

    Main Methods:

    • Pregnant rats received alcohol via intubation throughout gestation; controls received vehicle and were pair-fed.
    • Offspring were surrogate-fostered to non-treated dams at birth.
    • Behavioral testing (two-way shock avoidance task) and neuroanatomical analysis of the hippocampus were conducted at 11 weeks of age.

    Main Results:

    • Alcohol-exposed offspring performed significantly worse in the two-way shock avoidance task compared to controls.
    • Significant deficits in dendritic structure were observed in the hippocampus of alcohol-exposed animals.
    • A correlation was found between behavioral performance and neuroanatomical alterations.

    Conclusions:

    • Prenatal alcohol exposure leads to persistent learning and memory deficits in adult offspring.

    • These findings support the link between prenatal alcohol exposure and learning disabilities seen in Fetal Alcohol Spectrum Disorders.
