    Background:

    • Lung cancer diagnosis relies on accurate detection of tumor markers.

    Purpose of the Study:

    • To evaluate the efficacy of a rapid, sensitive thin layer immunoassay (TLI) using Visualization of Condensation on Plastic Surface (VCS) method.
    • To test monospecific and monoclonal antibody-derived lung tumor-associated antigens (TAA and TAA epitopes) for detecting lung cancer in human sera.
    • To assess the potential of VCS-TLI for aiding pathological evaluations and monitoring immunotherapy.

    Main Methods:

    • Performed 684 rapid, sensitive thin layer immunoassays (TLI) using an adapted Visualization of Condensation on Plastic Surface (VCS) method.
    • Tested squamous cell and adenocarcinoma monospecific TAA and monoclonal antibody-derived TAA epitopes against coded human test sera.
    • Utilized coded sera from normal individuals and patients with non-lung cancers as controls.

    Main Results:

    • Both TAA forms yielded positive tests in 94% of lung cancer sera of appropriate histological types.
    • TAA epitopes showed higher specificity; 5% of non-lung cancer sera tested positive for TAA epitopes.
    • No reactivity was observed with sera from normal individuals.
    • Indications of utility in aiding pathological evaluations and detecting precancerous conditions in asbestosis patients.

    Conclusions:

    • VCS-TLI is a rapid and sensitive method for detecting lung cancer-associated antigens.
    • The assay demonstrates high accuracy in identifying lung cancer sera and low false-positive rates.
    • VCS-TLI shows potential for clinical applications, including early detection, pathological evaluation, and monitoring TAA immunotherapy.
