    Background:

    • Concanavalin A (Con A) is a T-cell mitogen used to study lymphocyte activation.
    • Lymphocyte activation involves complex signaling pathways and cellular commitment.
    • Understanding factors that modulate lymphocyte activation is crucial for immune response studies.

    Purpose of the Study:

    • To investigate the effect of alpha-methyl-D-mannoside (MAM) and corticosteroids on concanavalin A-induced lymphocyte activation.
    • To examine the role of the commitment phenomenon in lymphocyte activation suppression.
    • To assess the impact of prostaglandin E2 (PGE2) on committed lymphocytes.

    Main Methods:

    • Human peripheral blood lymphocytes were cultured with concanavalin A for 48 hours.
    • Lymphocyte activation was measured by [3H]thymidine incorporation and rosette formation.
    • Alpha-methyl-D-mannoside (MAM) and corticosteroids were added at different time points.
