    Summary

    Lymphocyte blastogenesis in dogs with multicentric lymphosarcoma is often normal, but depressed responses indicate advanced disease (stages IV-B and V-B). Larger dogs (>25 kg) showed higher stage IV/V prevalence.

    Background:

    • Multicentric lymphosarcoma is a common cancer in dogs.
    • Assessing immune function, specifically lymphocyte blastogenesis, can provide insights into disease progression and prognosis.

    Purpose of the Study:

    • To evaluate mitogen-stimulated lymphocyte blastogenesis in dogs with multicentric lymphosarcoma.
    • To correlate blastogenesis results with clinical stage, body weight, and lymphoma subtype.

    Main Methods:

    • Lymphocytes were isolated from 37 dogs diagnosed with multicentric lymphosarcoma.
    • Mitogen-stimulated lymphocyte blastogenesis assays were performed.
    • Clinical staging, body weight, and histopathological subtype were recorded.

    Main Results:

    • Most dogs exhibited normal lymphocyte blastogenesis, irrespective of disease stage.
    • Depressed blastogenesis was predominantly observed in dogs with advanced clinical stages (IV-B and V-B).
    • Dogs >25 kg had a significantly higher incidence of stages IV or V disease (P=0.04) compared to dogs <15 kg.
    • Well-differentiated lymphoma was more frequently diagnosed at stage III than poorly differentiated lymphoma.

    Conclusions:

    • Mitogen-stimulated lymphocyte blastogenesis may not be a sensitive indicator for early-stage canine multicentric lymphosarcoma.
    • Severely depressed blastogenesis suggests advanced disease and warrants further investigation.
    • Body weight and lymphoma differentiation are important factors in disease staging and presentation.
