    Background:

    • Previous studies suggested the presence of a human globin-related sequence, psi beta 2, within the human beta globin gene cluster.
    • This putative pseudogene was identified based on hybridization experiments using a human fetal globin cDNA plasmid.

    Purpose of the Study:

    • To determine the complete nucleotide sequence of the DNA region where the psi beta 2 sequence was thought to exist.
    • To investigate the nature of the psi beta 2 sequence and clarify its relationship to globin genes.

    Main Methods:

    • DNA sequencing of a 3.4 kilobase pair region within the human beta globin gene cluster.
    • Hybridization studies using a poly (dA-dT)-tailed human fetal globin cDNA plasmid (pJW151).
    • Analysis of hybridization under low stringency conditions and in the presence of poly(rA).

    Main Results:

    • Complete nucleotide sequencing revealed no globin-related sequences in the targeted region.
    • The region contains several stretches of poly (dA-dT).
    • Hybridization to the cDNA plasmid was confirmed but abolished by poly(rA), indicating it was due to the poly (dA-dT) tails.

    • The psi beta 2 sequence is not a globin pseudogene.
    • The observed hybridization was an artifact caused by poly (dA-dT) sequences in the DNA region interacting with the poly (dA-dT) tails of the cDNA plasmid.
    • This finding resolves a discrepancy in previous research on the human beta globin gene cluster.
