Hybridization between T and B lymphoma cell lines

M J Taussig, A Holliman, L J Wright

    Immunology
    |January 1, 1980

    Summary

Hybridizing a thymoma cell line with a B lymphoma line revealed dominant T-cell traits in hybrids. This suggests B-cell gene suppression and T-cell marker gene derepression following cell fusion.

    Area of Science:

    • Immunology
    • Cell Biology
    • Genetics

    Background:

    • The AKR thymoma line BW 5147 and the B lymphoma line WEHI 231 were utilized.
    • WEHI 231 cells express immunoglobulin M (IgM).

    Purpose of the Study:

    • To investigate the potential for hybridizing T-cell and B-cell lines.
    • To determine the phenotypic characteristics of resulting hybrid cells.

    Main Methods:

    • Cell hybridization techniques were employed to fuse BW 5147 and WEHI 231 cell lines.
    • Flow cytometry or similar methods were used to analyze cell surface marker expression (e.g., IgM, Thy-1).

    Main Results:

    • Successful hybridization occurred between the thymoma and lymphoma lines.
    • T-cell characteristics, specifically Thy-1 expression, were dominant in the hybrid cells.
    • IgM expression, a B-cell marker, was suppressed in all analyzed hybrids.

    Conclusions:

    • The BW 5147 thymoma line can hybridize with cells of non-T-cell origin, such as B lymphoma cells.
    • Cell fusion can lead to the suppression of B-cell gene expression.
    • Genes associated with T-cell markers can be derepressed following fusion with thymoma cells.
