    Summary

    Immune cells synthesize and secrete ferritin, a protein involved in iron storage. This study found that immune cells produce ferritin with a higher proportion of H subunits, which are linked to certain tumors.

    Background:

    • Ferritin is a protein complex responsible for iron storage within cells.
    • The cellular origin and specific subunit composition of secreted ferritin, particularly in immune cells, remain incompletely understood.
    • Elevated levels of certain ferritin subunits have been observed in the serum of cancer patients.

    Purpose of the Study:

    • To investigate the de novo synthesis and secretion of ferritin by human immune cells.
    • To determine the subunit composition of ferritin produced by mononuclear cells and lymphoid cells from healthy individuals and Hodgkin's disease patients.

    Main Methods:

    • Analysis of mononuclear cells and selected T and non-T lymphoid cells from peripheral blood and spleens.
    • Radioactive labeling of cellular lysates and supernatants.
    • Immunoprecipitation using antiserum to human liver ferritin.
    • Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) to analyze ferritin subunits.

    Main Results:

    • Immune cells, including T cells, synthesize and secrete ferritin.
    • SDS-PAGE revealed two ferritin subunit bands (19,000 and 21,000 Da), corresponding to L and H subunits.
    • The H subunit (21,000 Da) exhibited higher radioactivity than the L subunit (19,000 Da).

    Conclusions:

    • Human immune cells contribute to the synthesis and secretion of ferritin.
    • The secreted ferritin molecule is characterized by a significant proportion of the H subunit.
    • These findings suggest a potential role for immune cell-derived ferritin in conditions associated with altered ferritin subunit levels.
