    Summary

    Various polyclonal lymphocyte activators (PLA) decrease B-memory cell function in mice. This selective reduction in B memory cells impairs immune responses without affecting T cells.

    Background:

    • Polyclonal lymphocyte activators (PLA) are used to study immune responses.
    • B memory cells are crucial for secondary immune responses.

    Purpose of the Study:

    • To investigate the effect of various PLA on B memory cell function in mice primed with sheep red blood cells (SRBC).

    Main Methods:

    • Mice were primed with SRBC and then injected with various PLA.
    • Spleen cells were analyzed for plaque-forming cell (PFC) responses and rosette-forming cells (RFC).
    • Cell fractions (B cell-rich and T cell-rich) were isolated for further analysis.

    Main Results:

    • PLA treatment decreased anti-SRBC PFC responses and the number of SRBC-specific B memory RFC.
    • The observed decrease in responsiveness was specific to the B cell-rich fraction.
    • No significant changes were observed in the T cell-rich fraction.

    Conclusions:

    • PLA selectively reduce B memory cell function by decreasing the number of B memory cells.
    • This effect occurs without inducing differentiation into end cells.
    • The potency of PLA in reducing B memory cell function varies.
