    Summary

This study explored noninvasive myelin imaging using lipophilic radioactive tracers. Results show iodobenzene distribution in the brain correlates with myelin, enabling white matter visualization.

    Area of Science:

    • Neuroimaging
    • Radiochemistry
    • Pharmacology

    Background:

    • Myelin imaging is crucial for diagnosing neurological disorders.
    • Current imaging techniques have limitations in specificity and resolution.
    • Developing noninvasive methods to visualize myelin is a significant research goal.

    Purpose of the Study:

    • To investigate the feasibility of noninvasive myelin imaging using lipophilic radioactive tracers.
    • To identify properties of ideal agents for myelin scanning, focusing on blood-brain barrier permeability, metabolism, and lipophilicity.
    • To correlate tracer brain distribution with myelin content.

    Main Methods:

    • Investigated properties of lipophilic tracers for brain imaging.
    • Correlated brain distribution of inert tracers with their octanol-saline partitioning.
    • Used iodine-125 labeled iodobenzene in rabbits for invasive studies (autoradiography/dissection).
    • Administered iodine-123 labeled iodobenzene to a monkey for tomographic imaging.

    Main Results:

    • Tracer brain distribution closely matched myelin distribution in rabbit studies.
    • Autoradiography and regional dissection confirmed the correlation.
    • Tomographic reconstruction in monkeys showed brain uptake patterns consistent with white matter distribution.
    • Iodobenzene demonstrated potential as a myelin-imaging agent.

    Conclusions:

    • Noninvasive myelin imaging using lipophilic radioactive tracers is feasible.
    • Tracer lipophilicity and blood-brain barrier permeability are key factors for effective myelin scanning.
    • Iodobenzene shows promise as a tracer for visualizing white matter noninvasively.
