    2'-Deoxycoformycin causes significant hemolysis in mice by destroying a sensitive cell subpopulation. This effect is transient and requires a 6-day recovery period before re-administration to avoid recurrence, indicating selective cell toxicity.

    Background:

    • 2'-Deoxycoformycin (dCF) is an adenosine deaminase inhibitor in clinical trials.
    • Known toxicities include lymphopenia, seizures, renal failure, and hemolysis.

    Purpose of the Study:

    • To investigate the mechanism and characteristics of dCF-induced hemolysis in a murine model.
    • To identify factors influencing the recurrence of hemolysis after dCF administration.

    Main Methods:

    • Mice were treated with dCF on various schedules.
    • Hemolysis duration and recurrence were monitored.
    • Splenectomy was performed in some animals.
    • ATP and dATP levels in erythrocytes were measured.
