    This study reveals that T lymphocytes regulate granulocyte-macrophage progenitor cell differentiation independently from proliferation. Depleting specific T cell subsets delayed differentiation, suggesting distinct regulatory pathways for these crucial blood cell development processes.

    Background:

    • Bone marrow cultures allow observation of granulocyte-macrophage progenitor cells.
    • Colony-stimulating factors (CSF/CSA) stimulate proliferation and differentiation.
    • Existing evidence suggests proliferation and differentiation are inseparable.

    Purpose of the Study:

    • To investigate whether proliferation and differentiation of progenitor cells are independently regulated.
    • To identify the role of T lymphocytes in regulating these processes.

    Main Methods:

    • Bone marrow and peripheral blood cells were cultured in semi-solid media.
    • T lymphocytes were depleted using complement lysis with specific monoclonal antibodies (OKT3/MBG6 and OKT11a).
    • Proliferation and differentiation were assessed using morphological, cytochemical, and functional criteria.
