    Summary

    Gold miners with suspected progressive systemic sclerosis (PSS) showed significant chromosomal abnormalities. These genetic changes, including unstable cells and aneuploidy, were similar to those in non-occupationally exposed PSS patients.

    Background:

    • An occupational link between progressive systemic sclerosis (PSS) and gold mining was identified in 1957.
    • South African gold miners are a population at risk for PSS.

    Purpose of the Study:

    • To investigate chromosomal aberrations in gold miners suspected of having PSS.
    • To compare chromosomal instability in gold miners with and without definite PSS.

    Main Methods:

    • Cytogenetic analysis of chromosomes from 18 gold miners.
    • Assessment of unstable cells (Cu cells), random aneuploidy, and sister chromatid exchanges.
    • Comparison with control subjects.

    Main Results:

    • Definite PSS cases showed a highly significant increase in Cu cells and aneuploidy (P < 0.001).
