    Background:

    • Prostacyclin (PGI2) is a vasodilator with known effects on coronary circulation.
    • Adenosine is a key mediator of coronary blood flow regulation.

    Purpose of the Study:

    • To investigate the influence of prostacyclin (PGI2) on adenosine release in isolated rabbit hearts.
    • To elucidate the role of adenosine in mediating PGI2-induced coronary vasodilation.

    Main Methods:

    • Isolated rabbit hearts were perfused using the Langendorff method.
    • Experiments involved varying concentrations of PGI2 and the adenosine antagonist aminophylline.
    • PGI2-biosynthesis inhibition was also employed.

    Main Results:

    • PGI2 dose-dependently increased coronary flow and myocardial adenosine release.
    • Low-concentration aminophylline (10 µg/ml) suppressed PGI2-enhanced flow and adenosine release.
    • Adenosine also increased coronary flow and PGI2 release, effects inhibited by low-concentration aminophylline.
    • High-concentration aminophylline (50 µg/ml) mimicked adenosine's effects on flow and PGI2 release.

    Conclusions:

    • PGI2-induced coronary vasodilation in isolated rabbit hearts is, in part, mediated by adenosine release.
    • Adenosine plays a significant role in the cardiovascular effects of PGI2.
