    Area of Science:

    • Dermatology
    • Pharmacology
    • Toxicology

    Context:

    • Investigating the local effects and systemic absorption of topical pharmaceutical agents.
    • Evaluating the safety and tissue response to prolonged application of prospidin ointment in a preclinical model.

    Purpose:

    • To determine the resorption and local tissue effects of 50% prospidin ointment applied long-term in rabbits.
    • To assess the reversibility of any observed adverse effects.

    Summary:

    • Long-term topical administration of 50% prospidin ointment in rabbits did not result in systemic absorption at tested doses (up to 100 mg/kg).
    • Localized necrotic changes were observed at the application site.
    • Complete epithelial regeneration, including hair regrowth, was documented within two months post-treatment cessation.

    Impact:

    • Provides crucial data on the localized toxicity and safety profile of prospidin ointment.
    • Informs clinical application guidelines and risk assessment for topical prospidin use.
    • Highlights the potential for complete tissue recovery after discontinuation.
