Effect of clonidine (Haemiton) on plasma renin level and angiotensin sensitivity in man
Insights
Spironolactone abolished clonidine
Area of Science:
- Pharmacology and Cardiovascular Science
Background:
- Clonidine is an antihypertensive agent.
- Spironolactone is a potassium-sparing diuretic and aldosterone antagonist.
- The interaction between clonidine, spironolactone, and the renin-angiotensin system is not fully understood.
Purpose of the Study:
- To investigate the combined effects of clonidine and spironolactone on renin activity.
- To assess the impact of these drugs on angiotensin sensitivity.
- To elucidate the mechanisms underlying clonidine's antihypertensive effects.
Main Methods:
- A study involving 15 participants.
- Administration of clonidine alone and in combination with spironolactone.
- Measurement of renin activity and angiotensin pressor dose.
Main Results:
- Clonidine significantly reduced renin levels and increased the angiotensin pressor dose.
- Spironolactone completely abolished the effects of clonidine on both renin activity and angiotensin sensitivity.
- This suggests opposing actions of the two drugs on the renin-angiotensin system.
Conclusions:
- Spironolactone counteracts clonidine's influence on renin activity and angiotensin sensitivity.
- The findings highlight the complex interplay between these medications and the renin-angiotensin system.
- Further research into alternative mechanisms of clonidine's antihypertensive action is warranted.
Abstract:
The effect of clonidine and of clonidine + spironolactone on renin activity and angiotensin sensitivity has been studied in 15 persons. Clonidine-induced angiotensin pressor dose and renin level reduction have been abolished by spironolactone. The phenomenon is explained by the contrary effects of the two drugs on renin activity and angiotensin sensitivity. Other possible mechanisms of antihypertensive clonidine effect are also discussed.
