    Summary

    Dexamethasone (DEX) increases systolic blood pressure in rats but does not appear to rely on sodium retention or renin-angiotensin system activation for this effect. Further research is needed to understand DEX-induced hypertension mechanisms.

    Background:

    • Glucocorticoids, such as dexamethasone (DEX), are known to affect blood pressure.
    • The precise mechanisms underlying DEX-induced hypertension require further elucidation.

    Purpose of the Study:

    • To investigate the impact of dexamethasone (DEX) on systolic blood pressure.
    • To assess the role of sodium balance and the renin-angiotensin system in DEX-induced hypertension.

    Main Methods:

    • Rats were administered dexamethasone (DEX) and monitored for changes in systolic blood pressure.
    • Urine volume and sodium excretion were measured.
    • Plasma renin substrate (PRS) and plasma aldosterone concentration (PAC) were analyzed.

    Main Results:

    • DEX significantly elevated systolic blood pressure within three days.
    • Concurrent saline administration did not enhance the blood pressure effects of DEX.
    • Urine volume increased significantly, with a slight tendency for increased urinary sodium excretion.
    • Plasma renin substrate (PRS) was significantly elevated on day 8, but plasma aldosterone concentration (PAC) remained unchanged.

    Conclusions:

    • DEX-induced hypertension in rats may not be primarily mediated by sodium retention.
    • Activation of the renin-angiotensin system does not appear to be the main driver of DEX-induced hypertension.
    • These findings suggest alternative pathways contributing to DEX-associated blood pressure elevation.
