    Summary

    This study developed fluorescent-antibody reagents to identify Cryptococcus neoformans serotypes. These reagents successfully typed the fungus in both cultures and fixed tissue samples.

    Background:

    • Four serotypes (A, B, C, D) of Cryptococcus neoformans are recognized.
    • Current agglutination tests are unsuitable for typing C. neoformans in fixed tissues.

    Purpose of the Study:

    • To create fluorescent-antibody reagents for C. neoformans serotyping.
    • To evaluate these reagents for typing the fungus in both cultures and fixed tissues.

    Main Methods:

    • Antisera were generated in rabbits against whole, formalin-killed C. neoformans strains of each serotype.
    • Antisera were labeled with fluorescein isothiocyanate and adsorbed with heterologous serotypes.
    • Adsorbed conjugates were tested against C. neoformans isolates and in paraffin-embedded tissue sections.

    Main Results:

    • Specific fluorescent-antibody conjugates differentiated between C. neoformans serotypes.
    • Seventeen out of 21 tested isolates were successfully serotyped in paraffin-embedded tissue sections.
    • The developed reagents showed efficacy in both culture and tissue-based typing.

    Conclusions:

    • Fluorescent-antibody reagents offer a viable method for serotyping C. neoformans.
    • This technique is applicable to fixed tissue samples, overcoming limitations of current methods.
    • The study provides a valuable tool for the identification and study of C. neoformans infections.
