    Background:

    • The physiological roles of pancreatic polypeptide (PP) remain incompletely understood.
    • Identifying target organs is key to elucidating PP's function.

    Purpose of the Study:

    • To identify the target organs of avian pancreatic polypeptide (aPP) in chickens.
    • To investigate the role of different aPP termini in receptor binding.

    Main Methods:

    • Radiolabeled aPP (125I-labeled at N- or C-terminus) was injected into fasted chickens.
    • Tissue samples from various organs were collected after perfusion and radioactivity was measured.
    • Competition assays using unlabeled aPP were performed to confirm receptor binding specificity.

    Main Results:

    • The N-terminal labeled aPP (aPP-N) disappeared faster from plasma than the C-terminal labeled aPP (aPP-C).
    • Significant binding of aPP-N was observed in the pancreas, spleen, duodenum, ileum, and bone marrow, reduced by unlabeled aPP.
    • Liver and proventriculus also showed reduced binding, though less marked.

    Conclusions:

    • The pancreas, spleen, duodenum, ileum, and bone marrow are identified as target organs for aPP in chickens.
    • The C-terminal region of aPP appears to be involved in its receptor binding.
    • Further investigation into aPP-C binding is warranted due to less conclusive results.
