    Background:

    • The pituitary gland is the primary source of human growth hormone (hGH).
    • The presence and function of hGH-like substances in peripheral tissues are not well understood.

    Purpose of the Study:

    • To investigate the presence and characteristics of hGH-like material in normal human tissues.
    • To differentiate between pituitary hGH and tissue-derived hGH-like material.

    Main Methods:

    • Radioimmunoassay (RIA) and receptor assays were used to quantify hGH-like material.
    • Sephadex G-100 chromatography and anti-hGH immunocolumns were employed for characterization.
    • Male rat liver membrane preparations were utilized for receptor binding assays.

    Main Results:

    • Extractable hGH-like material was detected in liver, kidney, lung, muscle, colon, stomach, and brain.
    • Liver tissue showed the highest concentration of hGH-like material (14.7 ng/g).
    • The tissue-derived material exhibited properties similar to pituitary hGH, including chromatographic elution and RIA parallelism, with a receptor to immunoassay potency ratio of 3.8:1.

    Conclusions:

    • Normal human tissues contain extractable hGH-like material.
    • This material shares characteristics with pituitary hGH, suggesting potential local functions.
    • Artifacts were rigorously excluded as a cause for the observed hGH-like material.
