A comparative study of free thyroxine estimations

H A Ross, J W Visser, P J der Kinderen

    Summary

Two commercial methods for measuring free thyroxine (FT4) in serum were evaluated. Both Immophase and Lepetit FT4 assays demonstrated good precision and correlated well with a reference method.

    Area of Science:

    • Clinical Chemistry
    • Endocrinology
    • Immunoassay Development

    Background:

    • Accurate measurement of free thyroxine (FT4) is crucial for diagnosing thyroid disorders.
    • Commercial immunoassays offer convenient alternatives to equilibrium dialysis for FT4 determination.

    Purpose of the Study:

    • To experimentally evaluate two commercial FT4 measurement kits: Immophase (Corning) and FT4 (Lepetit).
    • To assess the precision, accuracy, and reliability of these kits compared to a reference method.

    Main Methods:

    • Evaluation of two commercial FT4 assay kits (Immophase and Lepetit).
    • Assessment of assay precision (intra- and inter-assay coefficients of variation).
    • Comparison of results with a reference symmetric dialysis method.

    Main Results:

    • Both Immophase and Lepetit methods exhibited satisfactory ease of handling and rapidity.
    • Intra-assay coefficients of variation were ≤5% and inter-assay coefficients of variation were <8% for both methods.
    • High correlation coefficients (0.95 for Immophase, 0.93 for Lepetit) were observed when compared to the symmetric dialysis reference method.

    Conclusions:

    • The Immophase and Lepetit commercial FT4 assays are reliable and accurate for serum FT4 measurement.
    • These methods demonstrate good precision and correlate well with a reference technique, despite theoretical concerns about one kit.
    • The evaluated commercial kits provide a viable option for routine clinical laboratory use.
