    Background:

    • Chromosome studies in Hodgkin's disease are challenging due to low mitotic cell yield.
    • Previous research indicates difficulties in obtaining adequate cellular material for karyotyping.

    Purpose of the Study:

    • To investigate the feasibility and findings of chromosome studies in lymph node biopsies from Hodgkin's disease patients.
    • To compare the cytogenetic landscape of Hodgkin's disease with other lymphoma types.

    Main Methods:

    • Analysis of lymph node biopsies from 25 consecutive Hodgkin's disease patients.
    • Karyotyping and correlation with histological subtypes and treatment history.

    Main Results:

    • 12 of 25 samples lacked mitotic cells; 3 showed only normal cells.
    • 66% of unsuccessful samples were nodular sclerosing type; 58% of no-division samples were from previously treated patients.
    • Distinct aneuploid clones were found in only 4 of 10 patients with abnormal cells; others had single abnormal cells.

    Conclusions:

    • Hodgkin's disease exhibits a high failure rate in chromosome studies and a low incidence of clear aneuploid clones.
    • This contrasts sharply with other lymphomas, where 80% of samples yield mitotic cells representing aneuploid clones.
