    Summary

    Phospholipase C treatment of rat liver microsomes yielded diglyceride and altered vesicle morphology. Microsomal vesicles remained closed, retaining their bilayer structure after this enzymatic hydrolysis.

    Background:

    • Phospholipase C (PLC) is known to hydrolyze phospholipids.
    • Previous studies suggested PLC hydrolysis targets the outer leaflet of microsomal membrane bilayers.

    Purpose of the Study:

    • To investigate the products of phospholipase C hydrolysis in rat liver microsomes.
    • To examine the morphological effects of phospholipase C treatment on microsomal vesicles.

    Main Methods:

    • Treatment of rat liver microsomes (total, rough, smooth) with phospholipase C.
    • Analysis of hydrolysis products.
    • Morphological examination using electron microscopy and freeze-fracture techniques.

    Main Results:

    • Diglyceride was the sole hydrolysis product identified.
    • Microsomes retained vesicular structure, but vesicle diameter decreased (total/smooth by ~50%, rough by ~33%).
