Background:

The Poland anomaly is a rare congenital condition characterized by underdevelopment or absence of the pectoralis muscle and associated abnormalities.
Concerns have been raised regarding potential teratogenic effects of certain medications used during pregnancy, including Debendox.

Observation:

A case-control study examined antenatal drug exposure in 46 individuals with Poland anomaly and 32 with isolated pectoralis major absence.
Debendox (doxylamine-succinate, pyridoxine-hydrochloride) was prescribed in one case of Poland anomaly and one case of isolated pectoralis absence.

Findings:

In the cases reviewed, Debendox was not administered during the critical period of organogenesis.
No causal relationship could be established between Debendox exposure and the development of Poland anomaly or isolated pectoralis major absence in the 78 cases studied.
