    Background:

    • Estrogen plays a role in bone health.
    • The mechanism of estrogen's direct effect on bone cells is not fully understood.

    Purpose of the Study:

    • To investigate the presence of specific estrogen binding proteins (receptors) in bone cells.
    • To determine if the direct effects of estrogens on bone are mediated by specific estrogen receptors.

    Main Methods:

    • Cytosol preparations from various bone types (human, rat, rabbit) were incubated with radioactive estradiol.
    • Experiments included incubation with estradiol alone and in the presence of an antioestrogen (Nafoxidine).
    • Separation of bound and free estradiol was performed using dextran coated charcoal treatment and sucrose gradient ultracentrifugation.

    Main Results:

    • Specific estrogen receptors were not detected in the cytosol of any bone cells examined.
    • No evidence suggests that estrogen's direct action on bone is mediated by specific receptors.

    Conclusions:

    • The study failed to demonstrate the presence of specific estrogen receptors in bone cell cytosol.
    • The direct effects of estrogens on bone, if they exist, are likely not mediated through classical estrogen receptors.
